Newswise New Brunswick, N.J., August 28, 2014 The remodeling of a kitchen or bathroom changes the appearance of the room and improves its functionality. As investigators at Rutgers Cancer Institute of New Jersey and Rutgers and Princeton Universities have demonstrated, a similar concept comes into play in remodeling proteins found within a family of cancers that does not respond well to cancer-treating drugs -- cancers driven by what is known as activated Ras.
According to the National Cancer Institute, more than a third of all human cancers are driven by mutations in the Ras family of genes. When Ras is activated, it promotes tumor growth but also activates autophagy which helps to sustain that growth. These cancers remodel proteins using the cellular self-cannibalization process known as autophagy to capture and degrade intracellular proteins and protein-containing organelles. Since Ras-driven cancers often rely on autophagy for growth and survival, this raised the question: Is proteome remodeling by autophagy important, and if so, by what mechanism?
This question was answered in research published in the current online edition of Molecular Cell (doi: 10.1016/j.molcel.2014.07.019), by senior author Eileen White, PhD, associate director for basic science at the Cancer Institute of New Jersey, and colleagues. They compared the global proteome (all of the expressed proteins) of Ras-driven cancer cells where autophagy was present to those where autophagy was deficient.
In this latest study researchers found that autophagy affects a majority of the proteins in Ras-driven cancers, yet the process is highly selective as to which proteins are targeted. For instance, investigators found that autophagy eliminates proteins involved in non-essential pathways or those deemed toxic under stressful conditions. Meanwhile, autophagy-resistant proteins involved in pathways needed for stress survival and autophagy maintenance were spared. The authors say these sets of proteins can serve as biomarkers for monitoring autophagy in the clinical setting. The study also revealed that defects in the autophagy process caused accumulation of certain proteins that prompted activation of an immune response in cancer cells and led to cell death.
This remodeling process of the cell proteome by autophagy is an important immune-suppressive survival mechanism for Ras-driven cancers, and inhibiting autophagy can provide a means to target these aggressive cancers, notes White, who is also a distinguished professor of molecular biology and biochemistry at Rutgers School of Arts and Sciences.
Along with White, other authors on the study are: Robin Mathew and Sinan Khor, both Cancer Institute and Rutgers University; Sean Hackett, Princeton University; Joshua D. Rabinowitz, Princeton University and Cancer Institute; and David H. Perlman, Princeton University.

White kitchens bring a clean, light feel to any home, but many homeowners express concern about how to keep those cabinets looking pristine. In reality, the best way to maintain that sparkle is to never use your kitchen, but if you insist on actually cooking, you are likely to encounter yellowing cabinets, accumulations of grease, and stained and marred sinks and countertops. What fun.
The truth is, you dont need to be nuts to own an all-white kitchen. Just a little diligent, and armed with the tips below.

Why do those sparkling white cabinets start to yellow? The first culprit is exposure to direct sunlight, which tends to jaundice or fade painted wood (and laminated surfaces). You should consider draperies, blinds or window film to shield your cabinets from direct sunlight.
The second culprit is actually using your cooktop. As soon as you start cooking, those mouthwatering aromas rise into the air in the form of microscopic food or grease particles, and since they have to land somewhere, why not on your white cabinets? The result? Cabinets that turn yellow.
To prevent this, you cant let grease accumulate on your cabinets. As soon as you begin boiling, broiling or deep-frying, turn on the exhaust fan or range hood to filter out at least some of those minuscule bits before they reach the surrounding surfaces.
Writer Lorna Hordos suggests removing a yellow tinge this way: Wash your cupboards with a fizzy mixture of 1 cup vinegar, 2 cups warm water and 1 tablespoon baking soda. To keep cabinets white, give them a monthly bath with a solution of warm water and a grease-busting dish soap.
How to keep the rest of your white spaces looking great
Then there is the inevitable muck and grime that accumulate from oil and grease splatters. The folks at Kemper Distinctive Cabinetry remind you to not use abrasive cleansers or scouring pads on painted cabinets, as they can scratch or dull the finish. A soft cotton cloth dampened with warm water is usually sufficient.

Nearly finished on construction of a second Class A office building off Union Centre Boulevard, developer Schumacher Dugan Construction Inc. is now eyeing plans to build a third in the township.
At the end of 2012, Schumacher Dugan opened Union Centre Office Park I, a two-story, 32,000-square-foot office building at 9032 Union Centre Blvd. Tenants include Millikin & Fitton Law Firm, Liberty Mutual, Nationwide Insurance law offices, Employers Health and Re/Max Preferred Group.
That first $5 million building is about 70 percent leased presently, said Mark Schumacher, vice president of Schumacher Dugan.
Construction started earlier this year on a second building sharing the same parking lot, this time three stories and 48,000-square-feet at 9078 Union Centre Blvd. So-called Union Centre Office Park II, a $5 million to $6 million project, is slated to open in October, Schumacher said.
And the first tenant has been signed.
OfficeKey, which rents office space to executives and corporations, has signed a lease for about 12,000-square-feet on Office Park IIs third floor.
Once construction is completed, OfficeKey will relocate to the new space from its current location in the West Chester Twp. office building Centre Pointe at 9277 Centre Pointe Drive. OfficeKey manages five office spaces in greater Cincinnati, including West Chester.
OfficeKey provides a way for financial firms, law firms and branch offices to establish an office by subscribing rather than renting, said founder David Myers.
The traditional model is to rent space. The new way to do it is to subscribe to an office service, Myers said. You can work in any location, invite clients to any location, yet you can have a good location for your business.

New West Chester office building gets first tenant

Robert F. Maguire III, a prominent developer who helped shape L.A.'s skyline in the 1980s and '90s, is back.
Maguire led development of several of the city's best-known office buildings including U.S. Bank Tower, the tallest structure in Southern California. Now, at 79, Maguire says he is gearing up to develop again.
He plans to build an unconventional office at Playa Vista, the expansive former headquarters site of aviation titan Howard Hughes that has become a magnet for so-called creative firms such as YouTube.
The monolithic granite and glass towers Maguire built for corporate titans of the late 20th century are not attractive to technology, entertainment and digital media firms now leading the region's economic recovery, he says.
"Smart companies want innovative space," he said. "Conventional office space is going to have a hard time competing."
The Playa Vista office market is roaring, in part because the tech-centric Westside, sometimes known as Silicon Beach, is running out of space.
"Santa Monica is full," Maguire said. "The obvious replacement for major tenants is Playa Vista."
His plan is to build an office called WE3, which would be the third building at an office campus called Water's Edge at the intersection of Lincoln and Jefferson boulevards. Maguire has held part interest in Water's Edge since he built it in 2002.
The complex, about a mile from the Pacific Ocean, was intended from the start to be a campus for creative technology types, but the timing was off. The roaring tech boom of the late 1990s was over by the time Water's Edge hit the market and it sat empty until video-game giant Electronic Arts Inc. moved there in 2004.
EA ran into financial challenges and labored to sublease much of its space, but Water's Edge is now at more than 90% occupancy, Maguire said. He plans to sell the complex to an investor willing to become a partner in the development of WE3.

During the Aug. 12 and 19 meetings of the Development Permit Review committee (DPR) which analyses all discretionary permits in La Jolla outside La Jolla Shores for conformance to the La Jolla Community Plan DPR members considered the proposed, rooftop expansion of Georges at the Cove restaurant on Prospect Street.
The DPR submits its recommendations to the La Jolla Community Planning Association to be ratified/approved and submitted to the citys Development Services Department.
Restaurateur George Hauer is seeking a coastal development permit to convert an unused roof area atop the building he leases at 1250 Prospect St. to accommodate a 550-square-foot bar and reception area that would complement his existing dining terrace.
Although DPR members were satisfied that the project meets existing height and parking requirements, they asked that rooftop ventilation and air conditioning equipment visible from Prospect Street be screened from public view during the process, as proposed by La Jollan Gail Forbes during the Aug. 12 meeting (Forbes first made the request during the July meeting of the La Jolla Planned District Ordinance committee, which recommended approval of Hauers plans).
Hauer said he e-mailed Forbes telling her to hold my feet to the fire.
Thats why Im here, Forbes said Aug. 12, adding that the new waiting area and larger bar space would increase safety by preventing congestion in the existing bar and on stairs up to the restaurant patio.
To relocate the existing bar, additional equipment will need to be moved further toward the roofs edge along Prospect Street.
Architect Mark Steele, who is designing the addition, said he and Hauer hope to begin construction by December or January to avoid having to request a waiver for the citys moratorium on summer construction in the coastal zone.

Five roofers could be heard banging on the rooftop of Trinity Preschool in Merrill throughout the week, as they work to give them a brand new roof free of charge.
"We had to tear the whole roof off, tear all of the siding off so that we could flash everything correctly and now we're putting the shingles back up," said Ryan Ott, owner of Ryan Ott Development and Construction.
The preschool is the winner of the company's free roof giveaway, voted on by Lincoln County fair-goers. Twenty-seven families and non-profits wrote letters to Ott describing why they need a new roof. He then narrowed the group down to nine and let the people decide.
"Oh my gosh! We are so blessed," said Trinity Preschool Principal and Director Kathy Yahr. "It was just so exciting when Ryan called me and told me. I knew we were getting lots of votes. I was going to the fair everyday and people were saying 'I voted for you! I voted for you!' and then when he called we were all just so excited. Our whole congregation is celebrating."
The school was in rough shape.
"We had all kinds of problems, there were actually shingles that were loose, there was a lot of mold an mildue on the top of it, it was just an old decreped roof that was in much need of replacement," she said.
Ott donated their labor and ABC Supply and Atlas Shingles both provided the building materials. Ott said he wanted to help someone in need.
"I thought it would be a good way to give back to the community," he said. "There's a lot of people that need a new roof and it's an expense that can be pretty hard to come up with."
"It's been on our trustee's list of projects that need to be done for probably 5 years at least," said Yahr. "We're kind of in a place where if something is an immediate need, if a window is broken, we replace that. I don't know how long it would have been before it would have really happened. It might have been another five years and then we probably would have had more damage to the building."
It came just in time for the school year too, with a new crop of students coming in on Tuesday.

PRODUCT: Certain Honeywell ASCa Inc. emergency locator transmitters (ELTs) installed on various transport category airplanes. ACTION: Final rule; request for comments.
SUMMARY: The FAA is superseding Airworthiness Directive (AD) 2013-18-09 for the product listed above. 2013-18-09 required various one-time general visual inspections of the ELT transmitter units (TUs), and corrective actions if necessary.
This new AD corrects the manufacturer's name in the AD applicability and adds a source of approval for an installation. AD 2013-18-09 was prompted by a fire on a parked and unoccupied airplane; preliminary information indicated combustion in the area of the ELT TU. This new AD was prompted by the identification of an error in the applicability of AD 2013-18-09.
The FAA is issuing this AD to detect and correct discrepancies of the battery wiring installation inside the TU, which could result in an electrical short and possible ignition source.
DATES: This AD becomes effective September 9, 2014. Comments must be received by October 9, 2014.
