    Restaurateur seeks return to Groton site

    GROTON -- Ten years after his restaurant was torn down to accommodate improvements at the intersection of Route 119 and Sandy Pond Road, property owner Joseph Wong appeared before the Planning Board with plans to get back into food service.

    Wong, along with representative John Amaral and engineer Jesse Johnson, met with the Planning Board Thursday to unveil early plans for the development of Crossroads Plaza, the 788 Boston Road lot the restaurateur owns and which had been the site of the former Groton Jade Chinese restaurant.

    According to Johnson, the site plan presented at the pre-submission review included a pair of buildings, the larger of which would house a new 106-seat Chinese restaurant planned by Wong.

    Wong received approval from the board earlier this year for a pair of special permits originally awarded to him in 2006 and covering a more ambitious plan for the environmentally sensitive site.

    At the time, board members were told there were no immediate plans to develop the 4.5-acre corner site but that Wong wanted to keep his options open in case the economy improved.

    Wong first contemplated developing the site several years ago, when it became clear the state's plans to redesign the Four Corners intersection would require the demolition of his Groton Jade Chinese Restaurant.

    There was talk Wong would rebuild elsewhere on the site but as time went on, plans emerged for a full-scale redevelopment of the lot that included the construction of up to five buildings.

    But those early plans met resistance from town officials, including the Planning Board, which was unhappy with the number and size of the proposed buildings.

    In 2006, Wong changed his design team. He brought in engineering firm GPR Inc., which proceeded to make drastic alterations to the Crossroads concept including cutting the number of proposed buildings from five to four and reducing the size of the retail space from 43,000 to 19,000 square feet.

    On Thursday, Johnson told members the new plan now included only two buildings, the first of which would comprise 4,500 square feet and house a new restaurant.

    THE site of a former Hereford restaurant on the banks of the River Wye is finally set to be developed.

    Riverside Construction (Hereford) Ltd will start work in the New Year on 18 new luxury apartments within a secure gated complex off Greyfriars Avenue.

    The former Campions restaurant stood empty after it closed in the 1990s and was finally demolished in February 2013.

    The site has been the subject of a number of planning applications over the years but plans submitted by Paul Tobin, of Riverside Construction, were approved a year ago.

    Mr Tobin said that work will begin in the New Year and is expected to take around 15 to 18 months.

    The complex will feature a living green wall which is designed to look aesthetically pleasing, while reducing energy use, providing insulation and minimising carbon dioxide emissions.

    Hunters Estate Agents are handling sales and are accepting offers between 279,500 and 295,000.

    And the agents say they have already received substantial interest, with nine of the apartments already sold.

    Chromatin remodeling – Wikipedia, the free encyclopedia

    Chromatin remodeling is the dynamic modification of chromatin architecture to allow access of condensed genomic DNA to the regulatory transcription machinery proteins, and thereby control gene expression. Such remodeling is principally carried out by 1) covalent histone modifications by specific enzymes, i.e., histone acetyltransferases (HATs), deacetylases, methyltransferases, and kinases, and 2) ATP-dependent chromatin remodeling complexes which either move, eject or restructure nucleosomes.[1] Besides actively regulating gene expression, dynamic remodeling of chromatin imparts an epigenetic regulatory role in several key biological processes, e.g., DNA replication and repair; apoptosis; chromosome segregation as well as development and pluripotency. Aberrations in chromatin remodeling proteins are found to be associated with human diseases, including cancer. Targeting chromatin remodeling pathways is currently evolving as a major therapeutic strategy in the treatment of several cancers.

    The transcriptional regulation of the genome is controlled primarily at the preinitiation stage by binding of the core transcriptional machinery proteins (namely, RNA polymerase, transcription factors, and activators and repressors) to the core promoter sequence on the coding region of the DNA. However, DNA is tightly packaged in the nucleus with the help of packaging proteins, chiefly histone proteins to form repeating units of nucleosomes which further bundle together to form condensed chromatin structure. Such condensed structure occludes many DNA regulatory regions, not allowing them to interact with transcriptional machinery proteins and regulate gene expression. To overcome this issue and allow dynamic access to condensed DNA, a process known as chromatin remodeling alters nucleosome architecture to expose or hide regions of DNA for transcriptional regulation.

    By definition, chromatin remodeling is the enzyme-assisted process to facilitate access of nucleosomal DNA by remodeling the structure, composition and positioning of nucleosomes.

    Access to nucleosomal DNA is governed by two major classes of protein complexes:

    Specific protein complexes, known as histone-modifying complexes catalyze addition or removal of various chemical elements on histones. These enzymatic modifications include acetylation, methylation, phosphorylation, and ubiquitination and primarily occur at N-terminal histone tails. Such modifications affect the binding affinity between histones and DNA, and thus loosening and tightening the condensed DNA wrapped around histones, e.g., Methylation of specific lysine residues in H3 and H4 causes further condensation of DNA around histones, and thereby preventing binding of transcription factors to the DNA leading to gene repression. On contrary, histone acetylation relaxes chromatin condensation and exposes DNA for TF binding, leading to increase gene expression.[2]

    Well characterized modifications to histones include:[3]

    Both lysine and arginine residues are known to be methylated. Methylated lysines are the best understood marks of the histone code, as specific methylated lysine match well with gene expression states. Methylation of lysines H3K4 and H3K36 is correlated with transcriptional activation while demethylation of H3K4 is correlated with silencing of the genomic region. Methylation of lysines H3K9 and H3K27 is correlated with transcriptional repression.[4] Particularly, H3K9me3 is highly correlated with constitutive heterochromatin.[5]

    Acetylation tends to define the openness of chromatin as acetylated histones cannot pack as well together as deacetylated histones.

    However there are many more histone modifications, and sensitive mass spectrometry approaches have recently greatly expanded the catalog.[6]

    The histone code is a hypothesis that the transcription of genetic information encoded in DNA is in part regulated by chemical modifications to histone proteins, primarily on their unstructured ends. Together with similar modifications such as DNA methylation it is part of the epigenetic code.

    A Texas plumber says he has no clue how his old work truck wound up some 11,000 kilometres away in the hands of Islamic militants in Syria. But an extremist group, Ansar al-Deen Front, reportedly posted a photo of it on Twitter earlier this week, showing the black Ford F-250 with an anti-aircraft weapon in the bed.

    Since then, his company has apparently been getting bombarded with phone calls from people across the country, accusing it of aiding and abetting terrorists.

    The plumber'scompany logo and phone number were still printed on the driver-side door.

    "How it ended up in Syria, I'll never know," Mark Oberholtzer, who owns Mark-1 Plumbing in Texas City, told theGalveston County Daily News.

    His son, Jeff Oberholtzer, said the family had "no intentions or idea that this would happen." He added: "We have nothing to do with terror at all."

    Mark Oberholtzer told the Galveston newspaper he traded the truck in at AutoNation in Houstonlast year. He said he usually removes the decals beforehand, but figured the dealership would do it.

    "They were supposed to have done it and it looks like they didn't do it," he said.

    Trench collapse: Plumber dies at building site in Cherrybrook, in Sydney's north-west

    A plumber and father of two has been killed after falling into a collapsed trench at a building site in Sydney's north-west.

    Workers at the West Pennant Hills Sports Club desperately tried to dig the 47-year-old man out of the three-metre trench just after midday, but he died at the scene.

    Distraught colleagues were in tears at the worksite on Friday as Fire and Rescue workers tried to free the body.

    The ABC was told the man was the head of a family plumbing business and has two young daughters.

    The construction union said it was concerned safety standards are slipping at sites across the country, claiming workers are being rushed before Christmas.

    The CFMEU said four workers had died in New South Wales in the past fortnight.

    Police and Workcover are investigating the incident.
